What is this study?
The goal of this study is to evaluate the role of transcription factor EB (TFEB) in adipose (fat) tissue macrophages (ATM) in regulating adipose tissue and systemic metabolic function in obesity. The investigators will assess the differences in ATM lipid metabolism in people with metabolically abnormal obesity and lean individuals.

The goal of this study is to evaluate the role of transcription factor EB (TFEB) in adipose (fat) tissue macrophages (ATM) in regulating adipose tissue and systemic metabolic function in obesity. The investigators will assess the differences in ATM lipid metabolism in people with metabolically abnormal obesity and lean individuals. Both groups will have: * screening visit * imaging (body composition testing - dual-energy x-ray absorptiometry (DEXA) scans, magnetic resonance imaging \[MRI\] and magnetic resonance spectroscopy \[MRS\] scans) * Overnight visit with intravenous infusion (IV), muscle, and fat tissue biopsies

✅ You may be able to join if…
- •Be between 18 and 70 years old (inclusive of ≥18 and ≤70).
- •Not be pregnant or breastfeeding.
- •Be weight stable and mostly not do much exercise (sedentary) before joining.
- •Not use tobacco products or excessive alcohol, and not use supplements or medications that could change glucose or fat levels (except certain diabetes medicines in the MAO-type 2 diabetes group).
- •Have the right body fat and blood sugar/liver fat levels for one of the study groups (Lean, Metabolically Normal Obesity, Metabolically Abnormal Obesity, or MAO-type 2 Diabetes).
🚫 You may not be able to join if…
- •Be pregnant or breastfeeding.
- •Not meet all required study group inclusion rules (the exclusion list says anyone who does not meet all inclusion criteria is excluded).
- •Have significant organ system problems or diseases (example given: chronic severe kidney disease or cancer).
- •Use tobacco products, excessive alcohol, or supplements/medications that affect glucose and lipid metabolism (except certain diabetes medicines in the MAO-type 2 diabetes group).

What is this study testing?+

The investigators will assess the differences in ATM lipid metabolism in people with metabolically abnormal obesity and lean individuals.
Is it safe? Who makes sure of that?+
This is a research study. Every study is reviewed and monitored by an independent ethics board (called an IRB) whose job is to protect participants, and care is overseen by licensed medical staff. You'll be told the known risks before you agree to anything, and you can stop at any time.
Will I get a placebo instead of the real treatment?+

Do I have to stop taking my current medications?+
